چکیده

By B .iw ui BAIN, MAGDALENE R. VAS AND LoUIS LOWENSTEIN W HEN human peripheral blood leukocytes are cultured in vitro in the presence of phytohemagglutinin, some of the cells transform into immature forms that are capable of mitotic division.’ An incidental observation by Schrek and Donnelly2 indicated that, under certain conditions, a similar reaction might occur without phytohemagglutinin. These authors were studylug the morphology and motility of lymphocytes from 20 leukemic and nonleukemic patients, using the slide-chamber method and time-lapse cinematography. In one of their cultures, after 5 days’ incubation, they observed “a small number of large cells with large clear nuclei and prominent nucleoli.” A few of the cells were in mitosis. The authors noted that the culture “proved to be a mixture of bloods from 2 patients with hemochromatosis,” but did not elaborate on this observation. Since this one culture differed from the others only in that it was a mixture of leukocytes obtained from two subjects, it seemed desirable to us to determine whether the large cells would appear consistently when leukocytes from two normal individuals were mixed together and cultured in the absence of phytohemagglutinin.

Comparison of Several Maturation Inducing Factors in Dendritic Cell Differentiation

Background: Dendritic cells (DCs) are professional antigen presenting cells that have an important role in the initiation of immune response. The use of maturation factors in dendritic cell differentiation provides a promising approach in immunotherapy. Objective: In this study, we compared tumor necrosis factor-α, polyribocytidylic acid, lipopolysacharide and CpG oligonucleotides in inducing d...

Immediate Exposure to TNF-α Activates Dendritic Cells Derived from Non-Purified Cord Blood Mononuclear Cells

Background: Tumor necrosis factor alpha (TNF-α) is a primary mediator of immune regulation and might be required in the early stages of DC development from CD34+ cells. However, details of optimal timing of exposure to TNF-α in DC development process in monocytes or non-purified hematopoitic cells are still lacking and clear benefits of this approach to the development of DCs remain to be valid...

Comparison of Functional Competence of Umbilical Cord and Adult Peripheral Blood Dendritic Cells in Allogenic Mixed Leukocyte Reaction

Background: Dendritic cells (DCs) are the most potent stimulators of primary T cell responses and play a key role in immune reactions after stem cell transplantation. Very little is known about the cord blood (CB) dendritic cells and their potential involvement in the low incidence and lower severity of acute graft-versus-host disease after CB transplantation.   Objectives: The aim of this stud...

The Effect of Skin Homograft Rejection on Recipient and Donor Mixed Leukocyte Cultures

The lymphocyte proliferation in repeatedly studied mixed leukocyte cultures of peripheral white blood cells from a skin graft donor and 2 recipients was significantly increased at the time of graft rejection. This was determined from the increased proportions of mononuclear cells labeling with tritiated thymidine, increased mitotic indices, and the appearance of increased numbers of transformed...

The development of macrophages from large mononuclear cells in the blood of patients with inflammatory disease.

The origin and function of the increased of "atypical lymphocytes" which appear in the blood of patients with many inflammatory diseases is not known. Leukocyte suspensions from eight patients with systemic lupus erythematosus (SLE), five patients with other rheumatic diseases, and five patients with infectious diseases were pulse-labeled with tritiated thymidine (Tdr-(3)H) and sampled after 5 ...
